    摘 要:目的 对多种不同的噬菌体抗体库筛选方法进行比较研究. 方法 应用抗原固相化吸附筛选法、生物素化抗原液相筛选法和解离速率筛选法对半合成噬菌体抗体库或轻链替换库进行抗角蛋白噬菌体抗体的筛选，比较各自的筛选效率和优缺点. 结果 3种方法筛选抗角蛋白抗体均获成功，抗原固相化吸附筛选法效果可靠，所获抗体特异性高但抗原消耗量大;生物素化抗原液相筛选法方法敏感且节省抗原，可以根据实验目的和抗体库性质灵活调节筛选体系，其不足是筛选获得的克隆中容易出现非特异性结合的克隆;而解离速率筛选是选择性获得高亲和力抗体的有效手段. 结论 不同的筛选策略各具优势，在实际应用中可根据不同的实验目的选择相应的筛选方法.

    Abstract:AIM To study and to compare the efficiency of various panning strategies against phage antibody library.METHODS Three different methods including antigen-coat-ed solid phase selection，biotin-based liquid phase selection and the off-rate selection were applied to select anti-keratin phage antibodies from a semi-synthetic library and a light chain reshuffled library.Their advantages and disadvantages were analyzed.RESULTS Selections against keratin by the three methods were all successful.In comparison ......
