卡托普利对外源性羟自由基诱导心肌细胞凋亡的影响

关键词: 心肌细胞;凋亡;羟自由基;卡托普利摘 要:目的 观察卡托普利对外源性羟自由基诱导心肌细胞凋亡的影响. 方法 采用培养的第2代心肌细胞,随机分成4组:①正常对照组:仅用DMEM培养;②羟自由基组:OH-终浓度为10-4 mol L-1 ;③卡托普利组:卡托普利终浓度为10-5 mol L-1 ;④卡托普利+羟自由基组:卡托普利10-5 mol L-1 +10-4 mol L-1 OH-.观察心肌细胞存活率和形态学,流式细胞仪双标法检测细胞凋亡率. 结果 ①羟自由基组心肌细胞存活率降低,和对照组比较有显著性差异(P<0.05);卡托普利+羟自由基组细胞存活率较羟自由基组高(P<0.05).②Annexin V+ /PI- 细胞即凋亡细胞在羟自由基组有较高的发生率,明显高于其他各组(P<0.05);卡托普利+羟自由基组的细胞凋亡率显著低于羟自由基组(P<0.05).③羟自由基组凋亡心肌细胞呈特征性超微结构. 结论 卡托普利能减轻外源性羟自由基诱导的心肌细胞凋亡.

Abstract:AIM To observe effect of captopril on apoptosis of neonatal rat cardiomyocytes induced by exogenous hydrox-yl free radical.METHODS ①The second passage of cul-tured neonatal rat cardiomyocytes were divided randomly into four groups and were co-cultured with hydroxyl free radical(10-4 mol L-1 )and captopril(10-5 mol L-1 )in serum-free medium for six hours and24hours respectively.The survival ratio and morphology of cardiomyocytes and the percentage of Annexin V+ /PI- cells by flow cytometry were evaluated.RESULTS ①The survival ratio of cardiomyocytes was sig-nificantly decreased with10-4 mol L-1 hydroxyl free radical(P<0.05vs control)and significantly increased with both captopril(10-5 mol L-1 )and hydroxyl free radical.②The percentage of Annexin V+ /PI- cells with flow cytometry(apoptosis ratio)was increased significantly with10-4 mol L-1 hydroxyl free radical and decreased with both captopril and hydroxyl free radical(P<0.05).③Apopototic myocytes in hydroxyl free radical showed typical ultrastructures.CONCLUSION Captopril attenuates apoptosis of neonatal rat cardiomyocytes induced by exogenous hydroxyl free radi-cal. 0 引言 ......
