【摘要】 目的 探讨RhoA基因与肝细胞癌的发生、发展和侵袭、转移的关系。方法 采用逆转录聚合酶链反应(RT-PCR)在半定量水平检测42例肝细胞癌及其相对应的癌旁肝组织中RhoA mRNA的表达,同时采用PCR产物单链构象多态性(PCR-SSCP)银染法检测RhoA基因的突变情况。结果 本组42例肝癌组织中RhoA mRNA光密度相对值明显高于癌旁肝组织(P<0.01);有门静脉癌栓者其癌组织中RhoA mRNA光密度相对值明显高于无癌栓者(P<0.05)。经PCR-SSCP银染法分析未发现异常泳动条带。结论 RhoA基因在肝细胞肝癌进展中起重要作用,并可能与肝癌转移相关。【关键词】 肝癌;RhoA;基因表达;RT-PCR
Expression and its clinical significance of RhoA mRNA in hepatocellular carcinoma

【Abstract】 Objective To investigate the expression of RhoA gene in hepatocellular carcinoma and to evaluate the relationship between RhoA gene expression and invasion of hepatocellular carcinoma.Methods The mRNA expression of RhoA gene was examined by RT-PCR in 42 cases of hepatocellular carcinoma and adjacent non-cancerous tissue.In addition,the mutation of RhoA gene was examined by PCR-SSCP.Results The opacity density (OD) of RhoA mRNA expression in hepatocellular carcinoma tissues was significantly higher than that in adjacent non-cancerous tissues (P<0.01).The OD of RhoA mRNA expression in hepatocellular carcinoma with intrahepatic metastasis was significantly higher than that without intrahepatic metastasis (P<0.05).Mutation was not found in RhoA gene by PCR-SSCP examination.Conclusion The RhoA gene may be related to malignant transformation and development of hepatocellular carcinoma probably by overexpression. ......
