【摘要】 目的 观察过量碘化物对原代培养的猪甲状腺细胞凋亡的诱导作用,并对机制作初步探讨。方法 取健康的猪甲状腺细胞培养24h后,加入不同浓度的碘化钾(KI),观察细胞的形态改变;琼脂糖凝胶电泳、流式细胞术分析细胞DNA的降解;MDA法测定细胞膜的脂质过氧化程度。结果 加入KI 24h后,荧光染色镜下观察可见凋亡细胞;琼脂糖凝胶电泳呈梯状带;流式细胞术见亚二倍体(凋亡)峰,细胞凋亡率和脂质过氧化产物随碘化物浓度的升高而升高(P<0.05)。结论 过量碘化物诱导了原代培养的猪甲状腺细胞凋亡,可能通过活性氧机制启动凋亡。【关键词】 甲状腺细胞凋亡;过量碘;活性氧

【Abstract】 Objective To study the effect of excess iodide on the primary cultured pig and its mechnism. Methods We employed the primary cultured pig thyroid cells. The apoptosis was evaluated by AQ-stained fluorescin,DNA ladder by gel electrophoresis and subdiploidy by propidium iodide-stained flow cytometry. The level of lipid peroxide under KI treatment was determined by measuring the production of thiobarbituric acid reactive substances.Results Thyroid cells treated with iodide excess at different concentration underwent apoptosis. Iodide displayed a dose-dependent apoptosis index and a dose-dependent generation of TBARS levels.Conclusion Iodide excess induce the apoptosis in the primary cultured pig thyroid cells. This type of apoptosis seems to be activated by reactive oxygen species. ......
