     Effect of thermochemotherapy on multidrugresistant VX2 cell MDR gene

    【Abstract】 AIM: To observe the effect of thermochemotherapy on VX2 mdr1 cells MDR gene. METHODS: VX2 cells received hyperthermia (42.5℃ for 1 h), chemotherapy (ADM 32 μg/mL) and hyperthermia plus chemotherapy (42.5℃ for 1 h + ADM 32 μg/mL) respectively. The survival rate of cells and the expression of mdr1 mRNA and Pgp after thermochemotherapy were determined. RESULTS: The survival rate of the cells receiving hyperthermia plus chemotherapy was significantly lower than that of the cells receiving hyperthermia and chemotherapy respectively. After hyperthermia puls ADM, a lower (or lost) expression of mdr1 mRNA and lower expression of Pgp of VX2 mdr1 cells were found. CONCLUSION: Thermochemotherapy can decline tumor cell’s expression of mdr1 mRNA and Pgp, reverse tumor cell’s MDR, enhance the sensitivity of tumor cells to drugs. ......
