【Abstract】 AIM: To investigate the effects of EBV protein on the production of TNFα and IL8 cultured by human umbilical cord blood B cells. METHODS: B cells were isolated from umbilical blood, treated respectively with UV and heatinactivated EBV, and then they were cultured in complete IMDM. CD5, CD3, CD4 and CD8 cells were measured by flow cytometric analysis. TNFα and IL8 in the supernatant were detected by ELISA. RESULTS: No CD3, CD4 or CD8 cells were observed and CD5+ B cells account for 43% of all the detected cells at the 14th day, and 47% at the 28th day. Significant difference of the A value of TNFα in UVinactivated EBV group was observed at 2 d, 18 d, 22 d and 26 d time points (P<0.05) and significant difference of IL8 was observed 18 d, 22 d, 26 d and 30 d time points (P<0.05). But no significant difference of TNFα and IL8 A value was observed in heatinactivated EBV group(P>0.05). CONCLUSION: UVinactivated EBV is able to induce TNFα and IL8 production while heatinactivated EBV can not. The results suggest that EBV protein may exert some influence on the production of inflammatory factors. ......
