Simultaneous determination of Danshensu, protocatechuic aldehyde, paeoniflorin and ferulic acid in GuanXinⅡ decoction by RPHPLCGAN HongQuan, HUANG Xi, QU Yang, REN Ping, ZHANG ShanHong, MEI QiBing

【Abstract】 AIM: To establish a reversedphase high performance liquid chromatographic (RPHPLC) method for the simultaneous determination of Danshensu (Dhpl), protocatechuic aldehyde (Pal), paeoniflorin(Pa) and ferulic acid (FA) in GuanXinⅡ decoction. METHODS: The stationary phase was ZORBAX ExtendC18 reversedphase column (150×4.6 mm 5 μm), the mobile phase was A (MeOHHAC 99∶1, V/V)∶B (WaterHAC 99∶1, V/V) with linear gradient elution (0 min, A∶B=15∶85; 25 min, A∶B=40∶60) and the flow rate was 1 mL/min. Detection was performed at wavelength of 280 nm, 230 nm and 321 nm respectively for Dhpl, Pal, Pa and FA under room temperature. RESULTS: The linearity range of Dhpl, Pal, Pa and FA was 25.0-600.0 ng, 0.8-25.6 ng, 80.0-1280.0 ng and 10.0-160.0 ng and the correlation coefficient (r) of them was 0.9993, 0.9999, 0.9995 and 0.9994, respectively. The average recovery (n=5) was 102.19% for Dhpl (RSD=0.51%), 100.54% for Pal (RSD=4.67%), 99.99% for Pa (RSD=5.02%) and 98.6% for FA (RSD=6.72%). CONCLUSION: RPHPLC is a simple and effective method for quality control and therapeutic drug monitoring of GuanXinⅡ decoction. ......
