     Inhibitive effect of antisense oligonucleotide for cmyc on rat airway smooth muscle cell cycle

    【Abstract】 AIM: To observe the effects of antisense oligouncleotide for cmyc on rat airway smooth muscle cell cycle. METHODS: Rat airway smooth muscle cells were primarily cultured and lipofectin was used to introduce antisense oligonucleotide, sense oligonucleotide and mismatched oligonucleotide for cmyc into rat airway smooth muscle cells. Cell cycles were measured by flow cytometry and the expression of cyclin D1 and cyclin E was detected immunohistochemically. RESULTS: Antisense oligonucleotide for cmyc inhibited rat airway smooth muscle cells through S phase. Sense and mismatched oligonucleotides exerted no effect on cell cycles. Antisense oligonucleotide for cmyc decreased the expression of cyclin D1 and cyclin E. CONCLUSION: Antisense oligonucleotides for cmyc inhibit rat airway smooth muscle cells through S phase and decrease the expression of cyclin D1 and cyclin E. ......
