     Role of volumeregulated chloride channel in proliferation of human gastric carcinoma cell line SGC7901

    【Abstract】 AIM: To study the role of volumeregulated chloride channel in the proliferation of human gastric carcinoma cells. METHODS: Cell proliferation was analyzed by cell counting and MTT assay and the cell cycle analysis was carried out by flow cytometry. The currents of volumeregulated chloride channel were recorded by patch clamp. RESULTS: NPPB (100 μmol/L), a chloride channel blocker, reversibly decreased the cell number and the MTT optical density (OD) of SGC7901. Compared with control, NPPB arrested cells in G1 phase and extracelluar application of NPPB inhibited volumeregulated chloride currents which were induced by hypotonic stress. CONCLUSION: The volumeregulated chloride channel plays an important role in the proliferation of gastric carcinoma cell. ......
