     Comparison of cellcounting assays in cancer metastasis experiments in vitro

    【Abstract】 AIM: To compare the best cellcounting assay in cancer metastasis experiments. METHODS: Cultured cells with different density (1.1×102-2.2×105), the infiltrative cells in MatrigelBoyden chamber invasion assay and the adhesive cells in adhesion experiment were counted after being stained respectively by MTT crystal violet (CV) and HemateinEosin (HE). The results obtained by different staining methods were compared. RESULTS: The minimum detectable cell density was 2.75×104/mL and 4.3×102/mL respectively by MTT and CV staining methods. The CV staining method was fit in detecting the number of adhesive cells in adhesion experiment but unfit in detecting the infiltrative cells in MatrigelBoyden chamber invasion assay. The HE staining method was fit in testing the infiltrative cells. CONCLUSION: The HE staining method is good for the detection of infiltrative cells in MatrigelBoyden chamber invasion assay and CV method is good for the detection of adhesive cells. ......
