【Abstract】 AIM: To observe the effects of ectogenic nitric oxide (NO) on the differentiation of neural stem cells (NSCs) in vitro. METHODS: NSCs from mouse were cultured in medium containing EGF and on the fifth day, the medium of the experiment group was supplemented with sodium nitroprusside (SNP) as a donor of NO in vitro, while that of the control group was SNPfree. After 24 h, the cells were induced to differentiate in medium supplemented with serum and the differentiation of the two groups was observed. RESULTS: Cells in both groups differentiated into adult neural cells after cultured in medium supplemented with serum for 24 h, but the cells in the medium containing SNP differentiated earlier, with longer axons than those in the control. CONCLUSION: Nitric oxide can promote the differentiation of neural stem cells and the growth of neuritis of differentiated cells. ......
