【Abstract】 AIM: To investigate the association of paraoxonase 2 (PON2) Ser311Cys (S311C) gene polymorphism with type 2 diabetes mellitus (T2DM).METHODS: The S311C polymorphism in the PON2 gene was genotyped by Templatedirected Dyeterminator Incorporation with Fluorescence Polarization (TDIFP) in 17 patients with type 2 diabetes mellitus and 16 normal controls.RESULTS: In the 17 patients, there were 5 carriers of PON2 SS genotype, 6 of CS genotype, 6 of CC genotype; while in the 16 controls, there were 11 carriers of PON2 SS genotype, 3 of CS genotype, 2 of CC genotype. Patients with T2DM had a higher frequency of PON2 C311 allele than normal controls (P<0.01). C311 allele appeared to have an association with T2DM. The odds ratio was 4.018 with a 95% confidence interval of 1.371-11.774. CONCLUSION: TDIFP can detect PON2 S311C gene polymorphism correctly and sensitively. PON2 C311 allele may be a susceptible gene of T2DM. ......
