    Effects of tumor suppressor gene PTEN on human prostate cancer cell line PC3 in vitro

    【Abstract】AIM:To study the effects of tumor suppressor gene PTEN on human prostate cancer cell line PC3. METHODS: Cationic liposome LipofectAmine2000 was used to transfect retrovirus vector PbabePTENpuro containing PTEN and Pbabepuro into PTENdeficient prostate cancer cell line PC3 respectively, and then cell growth, adhesion, segregation, apoptosis, cell cycle and ultrastructure alteration in vitro were studied. RESULTS: After PTEN was transfected into PC3 and expressed, cell growth rate obviously slowed down and the growth inhibition rate was 83%. Adhesion rate dropped and cell segregated from matrix more easily. FCM showed a small apoptosis peak and an obvious G1 block in cell cycle and EMS showed that cells lost some malignant features in ultrastructure. CONCLUSION: PTEN can inhibit PC3 growth and adhesion in vitro and may play an important role in prostate cancer gene therapy. ......
