【Abstract】AIM:To study the effects of tumor suppressor gene PTEN on human prostate cancer cell line PC3. METHODS: Cationic liposome LipofectAmine2000 was used to transfect retrovirus vector PbabePTENpuro containing PTEN and Pbabepuro into PTENdeficient prostate cancer cell line PC3 respectively, and then cell growth, adhesion, segregation, apoptosis, cell cycle and ultrastructure alteration in vitro were studied. RESULTS: After PTEN was transfected into PC3 and expressed, cell growth rate obviously slowed down and the growth inhibition rate was 83%. Adhesion rate dropped and cell segregated from matrix more easily. FCM showed a small apoptosis peak and an obvious G1 block in cell cycle and EMS showed that cells lost some malignant features in ultrastructure. CONCLUSION: PTEN can inhibit PC3 growth and adhesion in vitro and may play an important role in prostate cancer gene therapy. ......
