    Isolation and purification of total saponins from cornus officinalis by column chromatography of macroreticular resin

    【Abstract】 AIM: To optimize the extraction and refinement of total saponins from Cornus officinalis by column chromatography of macroreticular resin. METHODS:Total saponins were extracted with 700 mL·L-1 of ethanol and purified with HPD 300 macroreticular resin and eluted with distilled water, 300 mL·L-1, 500 mL·L-1 and 700 mL·L-1 of ethanol. Adsorption capability and elution parameters were observed and compared with those extracted by routine solvents. RESULTS: Total saponins from Cornus officinalis enriched in 500 mL·L-1 of ethanol eluting solution. The dosage of elution reagents was 8 times as the sample volume and the elutive ratio was 818%. The average gaining rates of the two methods were 249% and 219% respectively. CONCLUSION: It is a good method to use macroreticular resin to adsorb and purify total saponins in cornus officinalis. ......
