     Expression of hypoxiainducible factor1alpha(HIF1α)and inducible nitric oxide synthase(iNOS)genes in inflammatory bowel disease

    【Abstract】 AIM: To investigate the expression and action of HIF1α and iNOS genes in inflammatory bowel diseases. METHODS: Paraffin embedded tissues from inflammatory bowel disease which had been diagnosed by clinical, endoscopic, histological and radiological examinations were used and immunohistochemistry method was performed to detect the expression of HIF1α and iNOS genes. RESULTS: The median of HIF1α positive expression rate was 54%, 59% and 1%, and that of iNOS expression was 58%, 54% and 2% in 47 ulcerative colitis, 13 Crohn’s disease and 10 normal colon mucosa respectively. The HIF1α and iNOS expression of ulcerative colitis and Crohn’s disease was significantly different from that of normal colon mucosa (P<0.01), while no difference was found between Crohn’s disease and normal colon mucosa (P>0.05). Linear correlation analysis showed that iNOS protein was positively correlated with HIF1α protein (ulcerative colitis: r=0.627; Crohn’s disease: r=0.610, P<0.05). CONCLUSION: HIF1α and iNOS are involved in the pathogenesis of inflammatory bowel diseases and HIF1α may be an essential regulator of iNOS gene expression. ......
