Influence of growth inhibition on HeLa cells by SBHL

    ¡¾Abstract ¡¿ AIM: To observe the inhibiting effects of SBHL on the growth of HeLa cells and to investigate the inhibiting mechanisms. METHODS: The proliferation of HeLa cells was observed by MTT assay and the changes of the cell cycle of HeLa cells treated with SBHL were analyzed by flow cytometry. The expressions of hTRT gene were detected by immunohistochemistry techniques and the differentiation of HeLa cells was observed by Wrightª²Giemsa staining. RESULTS: The proliferation of HeLa cells was inhibited at 0.1-20.0 mg/L SBHL. The inhibiting effect increased gradually following the increase of concentration of SBHL. The percentage of HeLa cells in S phase decreased and the percentage of cells in G1 phase increased after the cells were treated by SBHL, indicating that SBHL arrested HeLa cells in G1 phase and thus the synthesis of DNA was inhibited. After treatment by SBHL, the expressions of hTRT gene of HeLa cells decreased obviously. Differentiated HeLa cells wwere found after treatment by SBHL. CONCLUSION: ¢Ù The growth of HeLa cells is inhibited obviously by SBHL and the inhibiting effect is dependent on the concentration of SBHL. ¢Ú The expressions of hTRT gene are downregulated, the telomerase activity is inhibited and the growth of HeLa cell is inhibited. ......
