【Abstract】 AIM: To study the changes of intracellular [Ca2+] and cell activity in cortical neurons following ischemiclike conditioning and to explore the protective effects of dantrolene on the neurons. METHODS: Neurons were cultured and loaded with Flu3/AM, [Ca2+]i was measured by fluorescent intensity (FI) in each cell with confocal microscopy and the degree of injured neurons was analyzed with MTT reduction assay. RESULTS: After 10 min ischemiclike conditioning, a rapid increase in fluorescence intensity was observed. There was no difference between ischemic group and therapy group (P>0.05). The A value of injured neurons in ischemic group was significantly lower than that in therapy group. CONCLUSION: Dantrolene inhibits the increase of [Ca2+]i in cortical neurons and alleviates the degree of the injured neurons following ischemiclike conditioning. ......
