体外顺铂诱导肝癌SMMC┐7721细胞凋亡时细胞信号转导因子活性变化

关键词: 有丝分裂原激活的蛋白激酶家族;信号转导;细胞凋亡;顺铂;肝癌SMMC-7721细胞株;免疫细胞化学;流式细胞术摘 要:目的 体外使用顺铂诱导肝癌SMMC-7721细胞凋亡,检测细胞转导因子MAPKs及转录因子(JUN,CREB)活性的变化. 方法 采用免疫细胞化学法和流式细胞术法. 结果 15mg L-1 顺铂作用肝癌SMMC-7721培养细胞48h可观察到明显的凋亡峰,72h免疫细胞化学染色发现胞质或(和)胞核中MAPKs(JIN/SAPKs,p38MAPK,ERK)及转录因子(JUN,CREB)均有明显表达. 结论 在体外顺铂诱导肝癌SMMC-7721细胞凋亡过程中,MAPKs途径(JIN/SAPKs,p38MAPK,ERK)及转录因子(JUN,CREB)参与诱导了细胞凋亡.

Abstract:AIM To investigate activity changes of cell signal transduction factors(JIN/SAPKs,p38MAPK,ERK,JUN,CREB)in apoptotic hepatoma SMMC-7721cell induced by CDDP.METHODS Using immunocytochemistry and FCM methods.RESULTS After treatment with CDDP at15mg L-1 ,there were apoptotic peaks in cell cycle analysis on FCM for48h and expression of MAPKs and transcripton factors in cell plasm and/or nucleus for72h.CONCLUSION MAPKs pathway and transcription factors(JUN ......
