【Abstract】 AIM: To investigate the effect of antisense oligodeoxynucleotide bcl2 (bcl2 ASODN) on the expression of Bcl2 protein and the cell cycle of rabbit lens epithelial cells(LECs)induced by galactose. METHODS: From translation initiation region of bcl2 gene, oligonucleotides were synthesized and modified with phosphorothioate. Rabbit lens were treated with galactose and bcl2 ASODN. The influence of bcl2 ASODN on the level of Bcl2 protein expression and the cell cycle of LECs induced by galactose was detected by flow cytometry. RESULTS: 5.0-15.0μmol/L of bcl2 ASODN reduced the level of Bcl2 protein expression on LECs induced by galactose, with the fluorescence intensity decreasing from 158.70±2.14 to 113.94±2.88, and decreased the number of S phase cells of Lens from 20.60% to. 0.67%. CONCLUSION: bcl2 ASODN can inhibit the level of Bcl2 protein expression on LECs induced by galactose, interfere the entry of LECs into the S phase and inhibit the proliferation of LECs. ......
