【Abstract】 AIM: To investigate the effect of recombinant adenovirus encoding antisense cmyc (AdAscmyc) on the proliferation and cell cycle of HL60 cells and to study the molecular mechanism of the effects in vitro. METHODS: The HL60 cells were transfected with AdAscmyc and protamine sulfate. The growth of HL60 cells was observed by MTT assay and the changes of cell cycle were analyzed by flow cytometer in HL60 cells. The transcription of cmyc was assessed by RTPCR and immunocytochemical technique was used to test cmyc and proliferating cell nuclear antigen. RESULTS: The growth of HL60 cells was markedly inhibited by AdAscmyc. Flow cytometry analysis showed that the AdAscmyc transfection arrested HL60 cells in G0/G1 phase, reduced S phase and increased apoptosis rate. Transcription of cmyc was inhibited in the transfected HL60 cells. Immunocytochemical detection demonstrated that the AdAscmyc transfection suppressed the expression of cmyc and PCNA in HL60 cells. CONCLUSION: AdAscmyc can block cell cycle progression and inhibit the proliferation of HL60 cells in vitro. The biological effects of AdAscmyc may be associated with the expression of cmyc gene and PCNA. ......
