【Abstract】 AIM: To observe the effect of Gliclazide on mRNA expression of ATPsensitive potassium channels(KATP) in myocardium of diabetic rats. METHODS: Rat diabetic model was made by streptozotozin ip and fed on highfat diet for 6 weeks. The rats were then allocated randomly into 3 groups: the diabetes model group, the Gliclazide group and the positive control group(treated with insulin). A normal control group was also set up. KATPmRNA expression in myocardial cells of the rats was detected by the reverse transcriptasepolymerase chain reaction(RTPCR). RESULTS: There was no significant difference in mRNA expression levels of KATP in heart between diabetic, insulintreated diabetic and normal control groups(P>0.05). Comparison between Gliclazidetreated diabetic and nontreated diabetic rats showed that there was no change in mRNA levels of KATP in heart(P>0.05). CONCLUSION: The streptozotocininduced diabetes itself does not affect the mRNA expression of KATP(SUR2 and Kir6.2) in myocardial tissues. Gliclazide with treatment dosage has no effect on gene expression of KATP. ......
