Effect of Gliclazide on mRNA expression of ATPª²sensitive potassium channels in myocardium of diabetic rats

    ¡¾Abstract¡¿ AIM: To observe the effect of Gliclazide on mRNA expression of ATPª²sensitive potassium channels(KATP) in myocardium of diabetic rats. METHODS: Rat diabetic model was made by streptozotozin ip and fed on highª²fat diet for 6 weeks. The rats were then allocated randomly into 3 groups: the diabetes model group, the Gliclazide group and the positive control group(treated with insulin). A normal control group was also set up. KATPª²mRNA expression in myocardial cells of the rats was detected by the reverse transcriptaseª²polymerase chain reaction(RTª²PCR). RESULTS: There was no significant difference in mRNA expression levels of KATP in heart between diabetic, insulinª²treated diabetic and normal control groups(P>0.05). Comparison between Gliclazideª²treated diabetic and nonª²treated diabetic rats showed that there was no change in mRNA levels of KATP in heart(P>0.05). CONCLUSION: The streptozotocinª²induced diabetes itself does not affect the mRNA expression of KATP(SUR2 and Kir6.2) in myocardial tissues. Gliclazide with treatment dosage has no effect on gene expression of KATP. ......
