【Abstract】 AIM: To investigate the molecular mechanism involved in the islet formation and maturation of metabolic regulation by β cells. METHODS: The expression patterns of pancreas at embryonic day 15.5 (E15.5) and E18.5 were compared using the GeneChip Rat Expression Set 230 A. Based on the expression profiles of genes, the expression of mesothelin at different stages of rat pancreatic development was detected by RTPCR. RESULTS: Out of 1 319 genes of different expressions, the transcription factors and signal components related to cell differentiation were downregulated at the mRNA level while the genes related to islet formation and the function of metabolic regulation were upregulated. Mesothelin was specifically expressed at E18.5 pancreas, which was confirmed by RTPCR. CONCLUSION: It is from E15.5 to E18.5 that the islet is gradually formed and undergoes further remodeling and maturation. Mesothelin may be a novel regulator of the islet formation and maturation. ......
