【Abstract】 AIM: To study the characteristics of embryonic stem (ES) cells transfected with pTracer/mIFNγ. METHODS: The gene pTracer/mIFNγ including IFNγand green fluorescent protein (GFP) was transfected to obtain ES/ pTracer/mIFNγ cells by electroporation. RTPCR and ELISA were used to detect IFNγ expression at transcription and protein level respectively and green fluorescence was detected with fluorescence microscope to prove the GFP expression. Colony formation rates for ES/pTracer/mIFNγ cells and ES cell were conducted respectively. ES/pTracer/mIFNγ cells were induced to differentiate into neural precursor cells (NPCs) in serumfree selective medium and nestin immunoreactive was identified by immunocytochemistry. RESULTS: Green fluorescence was observed in ES/pTracer/mIFNγ cells and IFNγ concentration was (44.25±12.23)μg/L 72 h after gene transfection. ES/pTracer/mIFNγ cells were positive to alkaline phosphatase. The cell colony formation rates of ES cells and ES/pTracer/mIFNγ cells were 73.66% and 70.89%, respectively (P>0.05). ES/pTracer/mIFNγ cells differentiated into NPCs, which had potency to differentiate into all kinds of neural cells. CONCLUSION: The cells modified with IFNγgene may not influence the proliferation, differentiation and the pluripotentiality of ES cells. ......
