人细小病毒B19中国株VP1蛋白独特区基因片段序列变异分析

    摘 要:目的 研究B19病毒中国株独特区VP1的基因变异. 方法 采用聚合酶链反应技术(PCR)从两例中国再生障碍性贫血患儿血清中扩增VP1独特区基因片段,将其与PUC19连接,酶切鉴定筛选阳性克隆后测序分析. 结果 从一个患儿的血清中扩增出约480bp目的片段,并成功构建PUC19-VP1质粒,测序结果显示,与国外已发表的Wi株VP1独特区序列相比,有2处核苷酸发生改变,并可致所编码的氨基酸变化. 结论 中国B19病毒VP1独特区有变异.

    Abstract:AIM To obtain and sequence DNA clone of VP1unique region of human parvovirus B19from Chinese patients and to further study the genetic mutation of human par-vovirus B19of the Chinese strain.METHODS Two samples of VP1unique region of HPVB19were amplified from aplas-tic crisis serum of two Chinese children by PCR.The target fragment was linked with PUC19.Positive clones were cho-sen by identification of endonuclease cut and sequenced.RE┐SULTS A target fragment of480bp was amplified by PCR from the serum samples of one patient.Compared with the published B19VP1unique sequences ......
