关键词: 邻苯二甲酰亚胺;异鲁米诺衍生物;合成;化学发光;化学发光免疫分析摘 要:目的 建立一种合成化学发光标记试剂6-[N-(4-氨基丁基)-N-乙基]氨基-2,3-二氢-1,4-酞嗪二酮(ABEI)的简便、经济的新方法. 方法 室温下用KOH将邻苯二甲酰亚胺转化为邻苯二甲酰亚胺钾盐,并进一步进行甲基化、硝化和还原反应,得到N-甲基-4-氨基邻苯二甲酰亚胺.N-(4-溴丁基)邻苯二甲酰亚胺与N-甲基-4-氨基邻苯二甲酰亚胺缩合之后,通过乙基化和肼解反应得到目的产物ABEI. 结果 按起始原料计算,ABEI的总收率为4.4%.经熔点、核磁、质谱、红外、元素分析等方法鉴定,各中间体及目的产物均与其分子结构相符. 结论 起始原料价廉易得,反应步骤操作简便,有利于降低反应成本.
