【摘要】 目的 探讨还少丹对老年大鼠肝细胞老化改变的影响。方法 以24只健康SD老年大鼠为研究对象,用还少丹水煎液给药物组老年大鼠灌胃,用等量生理盐水给对照组大鼠灌胃,80天后断头处死大鼠,取鼠肝提取DNA,用碱变性法检测鼠肝DNA增色效应:用硫代巴比妥酸(thiobarbituric acid,TBA)法和荧光光度法分别测定鼠肝丙二醛(malondialdehyde,MDA)和脂褐素(lipofuscin,LF)含量。结果 还少丹能显著降低鼠肝碱变性增色效应(P<0.01);并能降低鼠肝丙二醛和脂褐素的含量,结果差异均具有非常显著性(P<0.01)。结论 (1)鼠肝DNA碱变性增色效应、丙二醛和脂褐素含量的改变,反映了肝细胞的老化过程;(2)还少丹具有拮抗鼠肝衰老的作用。【关键词】 抗衰老;碱变性;增色效应;丙二醛;脂褐素
Effects of huanshaodan on aging change in aging rats liver cells

【Abstract】 Objective To explore the effects of huanshaodan on aging change in aging rats liver cells.Methods Twenty-four healthy SD aging rats were used.The treatment group received gastric perfusion of Huanshaodan decoction,those in the control group were treated with gastric perfusion of saline of the same volume,for 80 days.The heads of rats were cut down for DNA obtained from the rats liver.The hyperchromic effects of DNA in the liver cell of old rats were detected with alkalis denaturation method.The contents of malondialdehyde and lipofuscin in the liver of old rats were detected with thiobarbituric acid and fluorospectrophometer analysis methods.Results Huanshaodan could not only remarkably decrease the hyperchromic effect of alkalis denatured DNA in the liver of old rats (P<0.01) ......
