环孢素A对大鼠脑缺血.再灌注损伤炎症反应的影响

[摘要]目的 探讨环孢素A对脑缺血.再灌注损伤的保护作用。 方法 将108只健康Wistar大鼠随机分为假手术组、生理盐水对照组和环孢素A组,参照Zea.Longa线栓法制备局灶性脑缺血.再灌注模型,在脑缺血2h再灌注22h和70h后,分别对各组各时间点大鼠的行为学评分,并对脑红四氮唑(TTC)染色结果、缺血区白介素1β(IL.1β)含量和髓过氧化物酶(MPO)活性进行测定和分析。 结果 在各时间点环孢素A组与生理盐水对照组相比,前者行为学评分优于后者(均P<0.05);环孢素A组脑梗死灶体积比生理盐水对照组明显缩小(均P<0.05);环孢素A组局灶性缺血.再灌注后脑组织中IL.1β的含量明显降低(均P<0.01);环孢素A可显著抑制MPO的活性(均P<0.01)。假手术组与上述两组相比,各项观察指标无明显改变。 结论 炎症反应参与脑缺血.再灌注损伤,环孢素A可显著降低缺血.再灌注后脑组织中IL.1β的含量、减轻缺血区内白细胞的浸润,对大鼠脑缺血.再灌注损伤具有明显的保护作用。[关键词] 环孢素A;脑缺血.再灌注;白介素1β;髓过氧化物酶;大鼠

Abstract:Objective To explore the protective effect of cyclosporin A on the cerebral ischemia-reperfusion-in-jury in rats.Methods One hundred and eight Wistar rats were randomly divided into3groups.The shamed-oper-ation group,normal saline control group and the cyclosporin A group.The focal cerebral ischemia-reperfusion-injury models were made by suture-occluded method.The score of behavior obstacle in rats were rated.TTC staining was used to calculate the infarct size.The activity of myeloperoxidease(MPO)and the content of interleukin.1β(IL.1β)were measured at22h and70h reperfusion following ischemia2h.Results Compared with normal saline control group ......
