草栽与段木栽培的灵芝活性成分的分离与鉴定

     【摘要】 目的 研究草栽和段木栽培灵芝的多糖性质和含量; 考察以草代木栽培灵芝可行性的研究。方法 提取、分离、逆相透析方法研究二者子实体多糖。应用气相色谱、凝胶渗透色谱法测定多糖的组成、摩尔比和相对分子质量。IR、1H-NMR 和13C-NMR 等光谱确定多糖肽化合物结构。结果 草栽灵芝多糖肽(GLPG) 和段木多糖肽(GLPW ) 的重均相对分子质量(Mw) 分别为5.13×105 和5.85×105; 黏均相对分子质量(Mη)分别为6.21×105和6.96×105,二者具有相同的相对分子质量分布及光谱数据。从光谱分析中看出各单糖间的苷键构型相似, 均以β-苷键为主; 糖肽部分均含有相同的16种氨基酸, 糖的组份为鼠李糖、木糖、果糖、半乳糖、葡萄糖等, GLPW 还含有微量甘露糖。结论 草栽灵芝与段木灵芝多糖肽的主要理化特性相似,为草栽灵芝的综合开发、节约木材、保护生态环境与药用提供了科学依据。

    Isolation and identification of active components of Ganoderma lucidum cultivated with grass and wood log (Extraction, purification and characterization of glycopeptide)

    【Abstract】 Objective To study the amount and properties of glycopeptide of Ganoder-ma lucidum ( Leyss.ex Fr. ) Kaist cultivated with grass ( GLPG ) and wood log ( GLPW ) ; to investigate the feasibility of substitution of grass for wood log to cultivate G. lucidum.Methods Glycopeptides were extracted and isolated from the f ruitbody of G. lucidum foll-owed by dialysis; themonosaccharid constituent and relative molecular weight of polysaccha-rides were examined by GC and gel filtration; structure of glycopeptide was determined by IR, 1H-NMR 和13C-NMR.Results Relative molecular weight s of GLPW and GLPG glycol-peptides were (M w ) 5.13×105 and 5.85×105, (Mη) 6.21×105 and 6.96×105, respect-tively; the glucosidic bonds of both glycopeptides wereβ-type; both glycopeptides consist 16 amino acids and Rha, Xyl, Fru, Gal and Glc ( there was trace amount of Man in GLPW).Conclusion There are less physical and chemical difference between GLPG and GLPW. The purification rate of GLPG is 2.8 times to GLPW. ......
