[摘要] 目的 探讨BCG对小鼠Th1/Th2细胞免疫反应的调节作用,研究BCG是否增强小鼠Th1型免疫反应。方法 将实验动物分为对照Ⅰ组、BCG14组、对照Ⅱ组、BCG42组。BCG14组、BCG42组给予BCG每次0.03 mL, 隔日1次腹部皮下注射,对照组注射相同剂量的生理盐水。第14天,将对照Ⅰ组、BCG14组分别处死取脾脏,BCG42组则继续行BCG免疫,至第42天对照Ⅱ组及BCG42组处死取脾脏。用ELISA法测定脾脏单个核细胞(MNCs)培养上清液中IL4、IL5、IFNγ含量。结果 BCG14组、BCG42组IFNγ水平较对照组明显增加(t=3.73、3.86,P<0.01)。BCG14组IFNγ水平与BCG42组比较差异无显著性意义(t=1.68, P>0.05)。其他两项指标与对照组比较差异无显著性(t=1.05~1.17, P>0.05)。结论 BCG能上调小鼠Th1型免疫反应。[关键词] BCG;白细胞介素4;白细胞介素5;干扰素Ⅱ型;小鼠,近交BALB C
EFFECTS OF BCG ON THE REGULATION OF Th CELL IMMUNITY

[ABSTRACT]ObjectiveTo investigate the effect of BCG on the regulation of Th1/Th2 balance in mice and explore whether BCG increased type Th1 immune response. MethodsThe experimental animals were divided into Control Group Ⅰ and BCG14 Group; Control Group Ⅱ and BCG42 Group. For BCG14 Group and BCG42 Group, a dose of BCG 0.03 mL was injected subcutaneously every other day and normal saline, same dose, for control groups. On day 14, those in Control Group Ⅰ and BCG14 Group were killed and spleens removed; those in BCG42 Group continued BCG immunity and killed on day 42, and the spleens removed. The levels of interleukin4, interleukin5 and interferonγ of mononuclear cells (MNCs) supernatant were measured by ELISA. Results IFNγ levels in BCG14 and BCG42 groups were significantly higher than those in the control group (t=3.73, 3.86;P<0.01). The difference of IFNγ levels of MNCs supernatant between BCG14 and BCG42 was not significant (t=1.68, P>0.05). As compared with control groups, the difference of the other two items showed no significance (t=1.05-1.17, P>0.05).ConclusionBCG immunization could upregulate Th1type immune response. ......
