【Abstract】 AIM: To investigate the transfection of liposome containing pSV2IFNβ and its effects on the induction of apoptosis of human glioma cell line SHG44. METHODS: MTT assay was used to measure the proliferation of SHG44 cells transfected by human interferonβ liposome containing pSV2IFNβ and the IFNβ expression in SHG44 cells was observed by indirect immunofluorescence staining. FCM Annexin was used to detect the apoptotic SHG44 cells after transfection with pSV2IFNβ and the morphological changes of SHG44 cells were detected by electron microscopy. RESULTS: The proliferation of SHG44 was obviously inhibited after transfection of human interferonβ liposome containing pSV2IFNβ and the inhibition rates were 16.7% and 32.7% respectively on d 4 and d 6 after transfection. The results of indirect immunofluorescence staining showed that the IFNβ expressions of SHG44 cells were obvious after transfection and the results of FCM showed that the apoptotic cell rate of the transfection group and control group were 66.8% and 19.8%, respectively. Significant differences were found between the two groups (P<0.01). Morphological changes of apoptosis were also found by electron microscopy. CONCLUSION: Human interferonβ gene liposome containing pSV2IFNβ can transfect human glioma cell line SHG44 and significantly induce the apoptosis of cells line SHG44. ......
