     Hepatoxicity of sodium valproate in rat primary cultured hepatocytes

    【Abstract】 AIM: To explore whether the VPA hepatotoxicity is related to dose (serum level), age or polytherapy. METHODS: We added different concentrations of VPA, VPAplusPB or VPAplusCBZ into hepatocytecultured fluid and observed their toxicity on hepatocytes in vitro. ALT, AST, LDH of the cultured fluid were determined using automatic biochemistry analysis apparatus. RESULTS: ① At the concentration of VPA >50 mg/L, the activation of ALT, AST, LDH dose dependently increased significantly in young and adult rat hepatocytecultured fluid. ② With the adding of VPA or VPAplusPB or VPAplusCBZ, no significant change was observed in the activation of ALT, AST, LDH of young rat hepatocytecultured fluid. CONCLUSION: ① VPA induces hepatotoxicity in a dose dependent manner. ② There is no difference of hepatocyte injury between young and adult rats. ③ VPAplusPB or VPAplusCBZ will not aggravate the hepatocyte injure. ......
