[摘要]目的 研究硫代磷酸化修饰的血管内皮生长因子(VEGF)反义寡核苷酸(ASODN)对慢性红白血病细胞K562VEGF表达的影响。 方法 将终浓度分别为10、20、30μmol·L -1 的VEGF ASODN和错义序列与K562细胞分别孵育24、48h，另设空白对照组。采用RT.PCR检测VEGF mRNA的表达，免疫组织化学方法检测VEGF蛋白的表达。 结果 VEGF ASODN对K562细胞VEGF表达有明显的抑制作用，与错义序列组和空白对照组比较有显著性差异(P<0.05)。错义序列组与空白对照组比较VEGF的表达无显著性差异(P>0.05)。 结论 VEGF ASODN可下调慢性红白血病细胞K562VEGF mRNA和蛋白的表达水平。

    The measurement of inhibiting effect of vascular endothelial growth factor(VEGF)antisense oligodeoxynucleotides on VEGF expressed in K562cell

    Abstract:Objective To study the effect of VEGF fully phosporothioated antisense oligodeoxynucleotide(VEGF ASODN)on VEGF expressed in K562cells in vitro.Methods K562cells were incubated with VEGF ASODN(eventual concentration as follows:10、20、30μmol·L -1 )or scrambled sequence，compared with negative control.The expression of VEGF mRNA was measured by semi-quantitative RT-PCR.The expression of VEGF pro-tein was measured by immunohistochemistry.Results The result showed that VEGF ASODN had obvious inhibitive effect on expression of VEGF mRNA in K562cells ......
