EGCG对人胃癌细胞增殖及其裸鼠移植瘤生长的抑制作用

【摘要】 目的 观察表没食子儿茶素没食子酸酯(EGCG)对体外培养人胃癌细胞系(SGC-7901)细胞的增殖抑制作用和体内抗人胃癌作用。方法 MTT法检测EGCG对人胃癌(SGC-7901)细胞株的生长抑制作用;以人胃癌细胞裸鼠移植瘤模型治疗实验,评价EGCG治疗人胃癌的有效性;通过TUNEL和免疫组化检测移植瘤细胞的凋亡情况。结果 在体外抗癌细胞增殖实验中EGCG对人胃癌SGC-7901细胞的IC50为58.60μg/ml。EGCG对人胃癌细胞株SGC-7901裸鼠移植瘤的抑瘤实验中5mg/(kg·d) EGCG作用20天的平均抑瘤率为34.51%,P<0.05;10mg/(kg·d)EGCG作用20天的平均抑瘤率为45.81%,P<0.01;20mg/(kg·d)EGCG作用20天的平均抑瘤率为54.64%,P<0.01。瘤细胞凋亡增加,TUNEL阳性细胞计数均值EGCG 5mg/kg、10mg/kg和20mg/kg组分别为2.59±2.81、4.3±2.58和5.96±2.13。结论 EGCG对人胃癌细胞株SGC-7901有不同程度的生长抑制作用,对人胃癌裸鼠移植瘤生长的抑制作用可能与肿瘤细胞凋亡增加有关。【关键词】 EGCG;裸鼠;人胃癌细胞株;抑制作用;凋亡
EGCG inhibition on growth of human gastric carcinoma cell SGC-7901 and xenograft in nude mice

【Abstract】 Objective To observe the effect of tea (-)-epigallocatechin-3-gallate (EGCG) on antiproliferative effect in human gastric carcinoma cell strain SGC-7901 and further explore the effect of EGCG on the human gastric carcinoma cells inoculated into BALB/C nude mice. Methods Antiproliferative effect of EGCG against human gastric carcinoma cell lines was tested by MTT method. Antitumor effect of EGCG was determined using the xenografts models of human gastric carcinoma cell SGC-7901 in nude mice. The apoptosis of the xenograft preparations was examined by using the terminal deoxynucleotidyl transferase mediated dUTP biotin end labeling of fragmented DNA (TUNEL) and immunohistochemical staining.Results The fifty percent inhibition concentration (IC50)of EGCG against SGC-7901 cell were calculated to be 58.68 μg/ml. Average inhibitory rates of EGCG against xenograft tumors of human gastric carcinoma in nude mice were 34.51% (P<0.05) ......
