【Abstract】 AIM: To modify the techniques of orthotopic renal transplantation in rats and to observe the acute allograft rejection of renal transplantation in Wistar rats. METHODS: Orthotopic renal transplantation in 46 Wistar rats which had received SD renal grafts was performed by using techniques improved in vascular anastomoses and the procurement of donor grafts. The 36 rats which survived more than 7 d after transplantation were randomly divided into the control group (n=18) and the therapy group (n=18). FK506 0.1 mg/(kg·d) was subcutaneously injected in therapy group daily for 1 week after operation. The acute allograft rejection of all renal transplanted rats was observed. RESULTS: In the 46 operations, 36 (78.3%) rats survived more than 7 d after transplantation and 10 rats died. The operation time was 2-2.5 h, including the warm ischemia time (5-10 min), the cold ischemia time (15-30 min), the time of vascular endside anastomosis for inferior vena cava (5.0-10.5 min), the endside arota anastomosis time (8.5-12.0 min). The control group all died 7-14 d after transplantation and the histopathologic examinations showed acute allograft rejection. But the FK506 therapy group survived more than 14 d. CONCLUSION: The improved techniques can increase the survival rate of renal transplanted rats. The acute allograft rejection frequently occurs in the 2 weeks after operation. FK506 therapy can effectively enhance the allograft survival rate after transplantation. ......
