普通低温与深低温处理的同种异体跟腱移植的免疫学实验研究

摘要: 目的:研究经普通低温(-26℃)、深低温(-80℃)、未经处理的同种异体跟腱和自体跟腱移植后的免疫学特点。方法:60只新西兰兔随机分为6组,A组经深低温冷冻处理的跟腱20条移植于C组,B组经普通低温冷冻处理的跟腱20条移植于D组,E组左右后肢跟腱互换移植,F组随机两两配对,然后将配对动物左右后肢未经处理的跟腱相互移植。未经处理的同种异体跟腱和自体跟腱切下后立即移植于相应受体,同种异体跟腱分别经普通低温冷冻、深低温冷冻处理10 d后才移植于相应受体。移植后4周采用补体依赖的淋巴细胞毒试验、腹腔巨噬细胞吞噬试验、淋巴细胞增殖试验测定各组细胞免疫学水平。结果:经深低温处理的同种异体跟腱移植后其淋巴细胞死亡率、增殖率和巨噬细胞吞噬率明显低于普通低温处理组(P<0.05),普通低温处理组又低于未经处理的同种异体跟腱移植组(P<0.05),而深低温处理组与自体组差异无统计学意义(P>0.05)。结论:经普通低温处理的异体跟腱移植后免疫原性强,而经深低温处理的异体跟腱移植后免疫原性弱,与自体跟腱移植后的免疫学特点基本一致。关键词:同种异体跟腱;移植;普通低温;深低温;免疫原性
The experimental study of transplantation immunology on rabbit tendon

Abstract: Objective: The characteristics of immunology were investigated after tendon allografts had been transplanted. Methods: NewZealand donor rabbits′ Achilles tendon auto grafts and fresh Achilles tendon allografts after cut off were immediately transplanted into corresponding acceptors. Donor rabbits′ Achilles tendon after cut off which were frozen to -26℃ and kept 10 days as ordinaryfrozen allgraft providers, donor rabbits′ Achilles tendon after cut off which were slowly frozen to -80℃ and kept 10 days as deepfrozen allgraft providers, after 10 days these two group providers were transplanted into corresponding acceptors. After transplanted four weeks functions of cellular immunity were measured through lymphocyte mortality test lymphocyte proliferating test and phagocytizing rate of celiac macrophages. Results: Lymphocyte mortality and lymphocyte proliferating rate and phagocytizing rate of macrophages in the deepfrozen group were lower than that in ordinaryfrozen group (P<0.05), in those were lower in ordinaryfrozen group than that in fresh allograft group (P<0.05), there was no statistical difference between the auto graft group and the deepfrozen group (P>0.05). Conclusions: Immunogenicity in ordinaryfrozen group was stronger both than that in deepfrozen group and the auto graft group. Immunogenicity in the deepfrozen group was quite lower, its characteristics of immunology after transplantation are similar to that in the auto graft group. ......
