摘要：目的研究鱼腥草挥发油脂质体肺靶向效果给药。方法酰化磷脂以逆相蒸发-搅拌超声法制备鱼腥草挥发油脂质体，过柱分离检测包封率，显微镜法观察形态，最后通过小鼠静脉注射给药0.5 h后，肺部提取药物，通过气质联用和抑菌圈实验比较脂质体与乳剂给药浓度。结果鱼腥草挥发油脂质体药物包封率达94.77%，鱼腥草挥发油脂质体呈双层结构，其平均粒径为97 nm，鱼腥草挥发油脂质体给药后肺脏提取液中未检出甲基正壬酮，而亚油酸乙酯质量分数为0.259%，鱼腥草挥发油乳剂给药时两者均无，鱼腥草挥发油脂质体给药后肺脏提取液抑菌圈直径可达1.2 mm，鱼腥草挥发油乳剂给药时抑菌圈最大仅为0.5 mm。结论以逆相蒸发-搅拌超声法制备鱼腥草挥发油纳米酰化磷脂脂质体肺靶向效果优于乳剂。关键词：酰化磷脂；鱼腥草挥发油脂质体；肺靶向；抑菌圈实验；气质联用
Preparation of Nano Volatile Oil of Houttuynia cordata Thunb. Liposome and Lung Targeting Effect

Abstract:ObjectiveThe lung targeting effect of the volatile oil of Houttuymia cordata Thunb. liposome was studied.MethodsThe nano volatile oil of Houttuymia cordata Thunb. liposome was made of acylated phosphatide with succinic anhydride and prepared by reverse phase evapoation and agitating-ultrasonic dispersion, the entrapment ratio of liposome was studied by Sephadex-G25 chromatography, and the appearance of liposome was observed by micrography, through tail intravenous injection, the small white mice were given the emulsion and the nano liposome, and the mice were killed after 0.5 h, the lungs were taken out and triturated, the drugs in lungs could be abstracted by solvent and the drug concentrations could be compared by GC/MS and a series of bactericidal tests.ResultsThe entrapment ratio of liposome was 94.77%, the volatile oil of houttuymia cordata Thunb. liposome has two layers, the average size was 97 nm, in abstracting liquor, with the liposome, 2-undecanone could not be found and the quality points of 9,12-octadecadienoic acid ethyl ester was 0.259%, but with the emulsion,2-undecanone and 9,12-octadecadienoic acid ethyl ester were both not be found, in bactericidal tests and with abstracting liquor, after given the liposome, the diameter of the bactericidal circles could get to 1.2 mm, however, after given the emulsion, the diameter of the biggist bactericidal circle was only 0.5 mm.ConclusionThe nano volatile oil of houttuymia cordata thunb liposome made of acylated phosphatide with succinic anhydride and prepared by reverse phase evapoation and agitating-ultrasonic dispersion was better than the emulsion in lung targeting. ......
