自制阳离子脂质体介导的体内外基因转移研究

     [摘要]目的:研究阳离子脂质体介导体内外基因转移的效率。方法:采用逆相蒸发法制备出包裹TNFα DNA的脂质体,将其与体外培养的靶细胞共育,或将其直接注射至小鼠腹腔内,检测TNF α的分泌水平。结果:TNF α基因转染NIH3T3细胞和腹腔细胞,10 d后均可检测到TNF α的分泌。结论:研制的脂质体不但能将目的基因转移至体外培养的靶细胞中,还可通过腹腔内注射直接将TNF α基因转移至腹腔细胞,稳定表达TNF α。

    Study on the in vitro and in vivo cytokine gene transfer mediated by selfmade cationic liposome

    Abstract: Objective:To investigate the efficiency of cationic liposome in mediating gene transfer. Methods: With reversephase evaporation, we prepared cationic liposome encapsulated TNFα DNA, which were either cocultured with those in vitro cultured target cells, or were directly injected into the abdominal cavity of mice. Then, TNFα secretion level was measured. Results: TNFα secretion could be detected in the NIH3T3 cells and the peritoneal cells transfected with TNFα genes, 10 days later. Conclusion: Results show that liposome can mediate cytokine gene transfer efficiently in vitro and in vivo, and TNFα can be expressed stably. ......
