自制阳离子脂质体介导的体内外基因转移研究

[摘要]目的:研究阳离子脂质体介导体内外基因转移的效率。方法:采用逆相蒸发法制备出包裹TNFα DNA的脂质体,将其与体外培养的靶细胞共育,或将其直接注射至小鼠腹腔内,检测TNF α的分泌水平。结果:TNF α基因转染NIH3T3细胞和腹腔细胞,10 d后均可检测到TNF α的分泌。结论:研制的脂质体不但能将目的基因转移至体外培养的靶细胞中,还可通过腹腔内注射直接将TNF α基因转移至腹腔细胞,稳定表达TNF α。[关键词] 脂质体;基因转移;肿瘤坏死因子α;基因治疗
Study on the in vitro and in vivo cytokine gene transfer mediated by selfmade cationic liposome

Abstract: Objective:To investigate the efficiency of cationic liposome in mediating gene transfer. Methods: With reversephase evaporation, we prepared cationic liposome encapsulated TNFα DNA, which were either cocultured with those in vitro cultured target cells, or were directly injected into the abdominal cavity of mice. Then, TNFα secretion level was measured. Results: TNFα secretion could be detected in the NIH3T3 cells and the peritoneal cells transfected with TNFα genes, 10 days later. Conclusion: Results show that liposome can mediate cytokine gene transfer efficiently in vitro and in vivo, and TNFα can be expressed stably. ......
