局灶性脑缺血大鼠学习记忆能力与海马齿状回nNOS表达的关系

[摘要]目的 观察局灶性脑缺血再灌注大鼠学习记忆与海马齿状回神经元型一氧化氮合酶(nNOS)表达的关系。 方法 大脑中动脉线栓法制作大鼠局灶性脑缺血再灌注模型,5.溴脱氧尿核苷(BrdU)标记分裂增殖细胞,应用Y型电迷宫监测大鼠学习记忆能力,免疫组化单标和双标技术检测大鼠海马齿状回BrdU阳性细胞数和nNOS的表达。 结果 局灶性脑缺血再灌注对学习记忆能力损害明显,但随着再灌注时间的延长学习记忆能力有较好恢复,再灌注28d恢复明显。海马齿状回BrdU阳性细胞增加,14d达高峰;再灌注后7d nNOS表达增强,28d达高峰,BrdU.nNOS双标细胞在14d达高峰。 结论 局灶性脑缺血再灌注可增强大鼠海马齿状回细胞的增殖能力,增殖细胞nNOS的表达对大鼠学习记忆能力恢复有促进作用。[关键词] 脑缺血;齿状回;学习记忆;5.溴脱氧尿核苷;一氧化氮合酶
