王学敏 江伟 周明 赵刚 杭燕南【摘要】 目的 观察急性出血坏死性胰腺炎(AHNP)后不同时间点胰腺和肺组织中核转录因子κB(NFκB)的DNA结合活性动态变化,以及胰腺炎后应用中度低温对NFκB活性的影响。 方法 第一部分实验采用牛磺胆酸钠逆行注射造成大鼠AHNP模型。大鼠随机分为正常对照组以及AHNP常温2、5和12 h组,用电泳迁移率改变法(EMSA)测定各组大鼠胰腺和肺组织中NFκB的活性。第二部分实验将大鼠随机分为假手术组、AHNP常温组和低温组,诱导AHNP后2 h和5 h测定各组胰腺和肺组织中NFκB的活性。结果 急性胰腺炎后大鼠胰腺和肺组织NFκB活性均持续增强,且各时间点胰腺组织NFκB活性显著强于肺组织(P均<0.01)。与AHNP常温组相比,低温组AHNP后2 h和5 h肺组织NFκB活性均显著降低(P均<0.01),而胰腺组织NFκB活性与常温组差异不明显。 结论 急性胰腺炎伴有胰腺和肺组织NFκB活性的显著增强。中度低温对急性胰腺炎后肺组织NFκB的激活有抑制作用。

【Abstract】 Objective To observe the dynamic changes in the DNA binding activity of nuclear factorκB (NFκB) in rat pancreatic and lung tissues in acute hemorrhagic necrotizing pancreatitis (AHNP), and investigate the effects of moderate hypothermia on AHNPinduced NFκB binding activity in a timedependent fashion. Methods A group of 24 rats was randomly divided into control group, 2, 5 or 12 hours groups after reproduction of AHNP. NFκB binding activity in pancreas and lung tissues was determined using electrophoretic mobility shift assay (EMSA). In another group of 36 rats were randomized into three groups with equal number: shamoperated rats which were kept under 3737.5 ℃ after laparotomy; AHNP rats with normothermia (3737.5 ℃); AHNP rats with moderate hypothermia (3233 ℃). Activity of NFκB was assessed at 2 or 5 hours after reproduction of pancreatitis. Results DNA binding activity of NFκB in pancreas and lung tissue of AHNP rats gradually increased after the development of pancreatitis, and NFκB binding activity was more prominent in pancreas than that in lung tissue at each time point (all P<.01). NFκB binding activity was inhibited at 2 and 5 hours of moderate hypothermia in lung tissue (P<0.01), but there was no difference in pancreatic tissue between normothermic and hypothermic rats at each time point. Conclusion These data suggest that the expression of NFκB is significantly increased after the development of AHNP. Moderate hypothermia could inhibit the expression of NFκB in lung tissue in acute pancreatitis. ......
