【Abstract】 AIM: To establish twodimensional gel electrophoresis (2DE) in research of laterally spreading tumor (LST) cell line proteome. METHODS: Immobilized pH gradient (IPG) gels with different pH ranges were employed and a series of steps of 2DE were optimized. RESULTS: Satisfactory 2DE maps were obtained. 1356±43 spots were isolated when pH 3-10 line IPG gels were employed, and 1285±51 and 989 spots were isolated when pH 4-7 IPG gels with 250 μg and 150 μg protein loading were employed. CONCLUSION: With optimization of the experimental conditions, 2DE of LST cell line has been successfully established.
