Screening and identification of antihTNFα monoclonal antibody recognized hTNFα epitopes from 7TMphage display peptide libraryZHANG Cun, ZHANG YingQi, YAN Zhen

【Abstract】 AIM: To screen out short peptide epitopes from 7TMphage display peptide library, which are capable of combining with antihTNFα monoclonal antibody. METHODS: The epitopes of human tumor necrosis factor were screened from 7TMphage display peptide library by using antihTNFα antibody as target protein and identified by sandwich ELISA and dotblot. RESULTS: After 3 rounds of screening, 15 of the 50 phage clones were identified as positive clones, which bound to the antihTNFα antibody. The amino acid sequences of the positive clones were WTFKMQP, WPFKMSP, WPYKMIP and WNYKMLP. CONCLUSION: The framework of the screened epitopes is similar and the epitopes can specifically bind to the antihTNFα antibody, which provides the basis for the development of short peptide vaccines. ......
