Correlation between expression of selectin and acute rejection in small bowel transplantation in rats

    ¡¾Abstract¡¿ AIM: To establish the models of acute rejection in small bowel transplantation in rats,to analyze the changes of selectin expression on protein and gene level during small bowel allograft rejection in rats and to explore their significance in small bowel allograft rejection in rats.METHODS:Heterotopic small bowel transplantation was performed with inbred rat F344/N and Wistar.All recipients were divided into four groups: group¢ñof Wistar,group¢òof F344/N¡úF344/N,group ¢óof F344/N¡úWistar/A,and group ¢ôof F344/N¡úWistar/A+FK506.Six rats were put to death on postª²operation day (POD)3,5 and 7 and the intestine was cut under sterile condition.All the samples were examined histologically and the expression of selectin was detected with immunohistochemistry technique.The intraª² graft mRNA expression of selectin was determined.RESULTS:Histological examination showed that mild acute rejection occurred on POD 3 in group ¢ó,moderate acute rejection on POD 5 and severe acute rejection on POD 7,while in group¢ò,no acute rejection was found.The histological evidence of group ¢ô indicated that FK506 could effectively control acute rejection.Pª²selectin,Eª²selectin and Lª²selectin were highly elevated in group ¢ó on POD 3,significantly higher than those of other groups (P<0.01).Eª²selectin and Lª²selectin were elevated with the aggravation of the acute rejection while Pª²selectin was not significantly elevated.But the expression of the selectin was a bit elevated in group ¢ò and group ¢ô after the operation though the expression in group ¢ô was lower than that in group ¢ò.CONCLUSION: Early expression of Pª²selectin in small bowel transplantation rejection can predict the occurrence of acute rejection at an early stage. ......
