摘要: 目的 探讨钙增敏剂MCI154对失血性休克大鼠血管反应性的影响。方法 实验分2部分:在体实验,42只Wistar大鼠随机分为正常组、休克组、MCI154(0.1、0.5、1.0、2.0 mg/kg)组,观察大鼠失血性休克后,MCI154对去甲肾上腺素(NE)升压反应的影响;离体实验,48只大鼠分为正常组、休克组、MCI154(10-7、10-6、10-5、10-4mol/L)组,取失血性休克大鼠肠系膜上动脉,利用离体血管环张力测定技术观察MCI154对NE诱导失血性休克大鼠肠系膜上动脉收缩反应性的影响。结果 在体实验,大鼠失血性休克2小时后,NE的升压作用显著下降,给MCI154后,NE的升压反应进一步降低,低于正常对照组和休克对照组。离体实验,与正常对照组相比,失血休克后血管环对NE的收缩反应性显著降低;不同浓度的MCI154预孵30分钟可使血管环的反应性进一步降低(P<0.01),且有剂量依赖性。结论 MCI154C能降低失血性休克大鼠全身或局部血管的反应性。关键词:出血;休克;钙增敏剂;血管反应性
Effects of a new calcium sensitizer MCI154 on vasoreactivity of hemorrhagic shock rats

Abstract: Objective To observe the effects of MCI154 on vascular reactivity following hemorrhagic shock(HS) in rats.Methods The experiments were conducted in vivo and in vitro.In vivo,42 Wistar rats were randomly divide into 6 groups:normol control,hemorrhagic shock control and shock with MCI154(0.1、0.5、1.0、2.0mg/kg).The hemorrhagic shock(30mmHg for 2 hours) model of rat was adopted to observe the effect of MCI154 on pressor effect of norepinephrine(NE) in vivo.In vitro,48 rats were divide into 6 groups:normol control,hemorrhagic shock control and shock with MCI154(10-7、10-6、10-5、10-4mol/L) groups.Superior mesenteric artery(SMA) was used to observe the effect of MCI154 on NE inducedvascular contraction with isolated organ perfusion system.Results In vivo,the NEinduced pressor response following hemorrhagic shock(2 hours) was significantly(P<0.01) decreased.After pretreatment with MCI154,the pressor response of NE was further decreased,it was significantly lower than that of the normal and shock control group.In vitro,the contractile response of SMA to NE following hemorrhagic shock was significantly reduced as compared with the control group.After MCI154 pretreatment for 30 minutes,the responsiveness of SMA to NE was further decreased as compared to shock group(P<0.01),this effect of MCI154 was in dosedependent manner.Conclusion MCI154 can reduce the vascular reactivity of hemorrhagic shock rats including the systemic responsiveness and local vascular reactivity. ......
