     Expression of Na+/Ca2+ exchanger in liver tissues of rats with hepatic fibrosis

    【Abstract】 AIM: To observe the expression level of Na+/Ca2+ exchanger mRNA in rats with hepatic fibrosis induced by CCl4,to investigate its relationship with collagen metabolism and to explore its diagnosis value for hepatic fibrosis.METHODS: Hepatic fibrosis rat model was induced by subcutaneous injection of CCl4.The expressions of Na+/Ca2+ exchanger mRNA and α1(Ⅰ) procollagen mRNA were determined by RTPCR in situ.The content of hydroxyproline in liver tissue was detected with Jamall's method and the concentration of HA,CⅣ and PCⅢ in serum was measured with RIA method.The deposition of collagen in the liver tissue was observed with HE,VG and Masson staining.RESULTS: The expressions of Na+/Ca2+ exchanger mRNA and α1(Ⅰ) procollagen mRNA in the liver of rats with hepatic fibrosis increased obviously (semiquantitative score 245 vs 010,P<005).Also significantly increased were the content of hydroxyproline in liver tissue and the concentration of HA [(45579±11355) vs (11241±4562) μg/L,P<001],CⅣ [(15962±2964) vs (3569±968) μg/L,P<001] and PCⅢ [(26554±9821) vs (8999±1085) μg/L,P<001] in serum of rats with liver fibrosis.The change of these parameters was parallel to the pathological damage of the liver (r = +048,+064,+052,+049,+056 and +060 respectively,P<005).CONCLUSION: Na+/Ca2+ exchanger can be a useful index for the diagnosis of hepatic fibrosis,and further study is needed. ......
