β聚糖酶的诱导合成Ⅱ:初始pH值与培养温度的影响

摘 要 以绿色木霉(Trichoderma viride)LH-011为产酶菌株,利用自制复合碳源,对初始pH值、培养温度等因素对β聚糖酶合成的影响进行了研究。结果表明:以预处理玉米芯为碳源(浓度15 g/L),调节碳氮比为6.4,初始pH 4.8,150 r/min摇瓶培养,前期(32±1) ℃培养24 h,后期(28±1) ℃变温调控培养72 h,纤维素酶活力和木聚糖酶活力分别达到了3.76 U/mL和13.95 U/mL。关键词 纤维素酶; 木聚糖酶; 绿色木霉

Abstract The effects of initial pH value and culture temperature on βglycanase synthesis by Trichoderma viride LH-011 were studied .15 g/L pretreated corn cob was used as the main corbon source and inducer,mixed (NH4)2SO4 and urea and peptone as nitrogen source. C/N was regulated to 6.4,with the initial pH value of 4.8,at 150 r/min.The temperature was(32±1) ℃ in the first 24 hours and (28±1) ℃ for 48 h. The cellulase and xylanase activity reached 3.76 U/mL and 13.95 U/mL respectively after 72 hours cultivation. ......
