【Abstract】 AIM: To the study doseeffect relationship between the DNA damage of HepG2 cells and the effect of different concentrations of hydrogen peroxide (H2O2) for 2 h respectively, and to observe the protective function of lipoic acid from the DNA damage of cells at the same time. METHODS: HepG2 cells were divided into control group, damage group and protective group, then were added with different concentrations of H2O2 and lipoic acid. Two hours later, the single cell gel electrophoresis (SCGE) was used to detect the DNA damage. RESULTS: The DNA damage of cells was more serious with H2O2 concentration increasing. The tail length, the tail DNA content and the tail moment significantly increased (P<0.05). Lipoic acid could significantly decrease the tail length, the tail DNA content and the tail moment of damaged cells (P<0.05). CONCLUSION: H2O2 causes DNA damage of cells in a doseeffect manner, while lipoic acid can reduce effectively this DNA damage caused by H2O2. ......
