葡聚糖微型凝胶柱测定辣椒碱柔性脂质体包封率的条件探讨

     摘 要 目的 研究葡聚糖微型凝胶柱测定辣椒碱柔性脂质体包封率的操作条件。方法 以洗脱液的紫外吸收值为指标,比较葡聚糖微型凝胶柱中的水分、离心机转速、离心时间对包封及未包封药物洗脱速度的影响。结果 葡聚糖微柱采用1000 r/min,6 s离心除去3 mL的水后,加入的脂质体2000 r/min,5 min离心,一次可将脂质体泡囊(包封的辣椒碱)完全洗脱,而游离辣椒碱不被洗脱。结论 采用葡聚糖微型凝胶柱测定脂质体包封率,当控制水分、离心机转速、离心时间,不仅速度快,而且葡聚糖用量少。

    Optimized separation conditions for the determination of encapsulation efficiency of liposomeencapsulated capsaicin

    Abstract Objective To optimize the separation conditions for the detection of encapsulation efficiency of liposomeencapsulated capsaicin on Sephadex G50 minicolumn. Method The effects of water content of the gel, centrifuge speed and time on the measurement of encapsulation efficiency were investigated using UV absorbance of the eluate as standard. Result When Sephadex G50 was centrifuged at 1 000 r/min for 6 s for removing 3 ml of water, then at 2 000 r/min for 5 min, liposomeencapsulated capsaicin was separated thoroughly from the gel and free capsaicin remained in the column. Conclusion Proper water content of the gel, suitable centrifuge speed and time are helpful for the separation of liposomeencapsulated capsaicin from the free capsaicin on the column. ......
