摘要:目的与方法 通过探讨用MTT法检测重组人表皮生长因子(rhEGF)复方抗菌凝胶生物活性的试验参数,包括凝胶剂中卡波姆含量对测定的屏蔽作用、不同试剂对卡波姆组分屏蔽作用的解除、抗菌成分磺胺嘧啶银浓度以及EGF含量对复方凝胶检测方法的影响作用等,建立一套适合用于EGF复方抗菌凝胶剂生物活性的检测方法。结果 优选出的最佳条件:EGF质量分数为20 μg/g,卡波姆质量分数为1.0%,用尿素+PBS+甘氨酸作屏蔽剂,磺胺嘧啶分数浓度为0.5%~2.0%。结论 本文方法可快速准确地对EGF复方凝胶活性进行检测,从而为复方凝胶剂的开发提供条件。关键词:重组人表皮生长因子;磺胺嘧啶银; 复方 ;凝胶剂 MTT法

Abstract: Objective To study several experiment data for the bioactivity assay of rhEGF gel with MTT method, including the masking effect of measuring by the quantity of carbomer, the eliminating masking effect by different reagents, the effect of SDAg and EGF concentrations on the detecting method of gel, we develop a new modified MTT method for the bioactivity assay for the compound rhEGF SDAg gel. Results The experimental result indicates that the assaying method of processing of gel samples with Urea+PBS+Gly, and assaying the filtrate with modified MTT and enzyme linked immunosorbent assay (ELISA) is applicable in the bioactivity assay of the compound gel, contribute to the development of a new rhEGF gel with antimicrobial ingredients with its quick and accurate advantage. ......
