摘要:目的 分析Monascus ruber 5031洛伐他汀合成酶基因的结构。方法 用3对以土曲霉洛伐他汀合成酶基因为基础设计的PCR引物对红曲霉基因组进行了PCR分析,其中一对引物序列为,正向:5’TTC GAT GCG GCC TTC TTC AAT3’和反向5’ATG GTT CGG CAT CGT AAT TCC 3’ 。结果 在红曲霉基因组中相当于土曲霉洛伐他汀合成酶基因的LNKS区域扩增出了745 bp的核酸片段,其序列与土曲霉合成酶的这一区域序列相同。结论 土曲霉和红曲霉洛伐他汀合成酶基因存在同源区域。关键词:红曲霉平;土曲霉;洛伐他汀合成酶合成酶;PCR
PCR analyses of Monascus ruber 5031 monacolin K biosynthetic gene cluster

Abstract: Objective Analyses the structure of Monascus ruber 5031 lovastatin biosynthesis gene cluster. Methods Three pairs of degenerate PCR primers were designed matching the regions of lovastatin biosynthesis gene cluster in Aspergillus terreus,one of them is,5’primer: TTC GAT GCG GCC TTC TTC AAT,3’primer: ATG GTT CGG CAT CGT AAT TCC. Results A 745bp DNA sequence was amplified by PCR from Monascus ruber 5031 genomic DNA and the sequence is the same with Aspergillus terreus lovastatin biosynthesis gene. Conclusion There is similar sequence within the lovastatin biosynthesis gene cluster of Aspergillus terreus and Monascus Ruber. ......
