阻断ckit对小鼠小肠Cajal间质细胞的影响及机制初探

摘要 目的:观察阻断ckit对小鼠小肠Cajal间质细胞(ICC)的影响以及与Cajal间质细胞ckit基因表达的关系。方法:选取新出生Balb/c小鼠,腹腔内隔天注射ckit抗体(ACK2)100 μg,共5次,对照组注射生理盐水,于小鼠出生后第10天观察小肠Cajal间质细胞的分布情况并检测小肠肌条ckit mRNA及蛋白的表达。结果:阻断ckit后小鼠小肠中未能明显观察到ICC分布,并且ckit mRNA表达以及ckit蛋白表达明显减少。结论:阻断ckit可导致小鼠小肠Cajal间质细胞缺失,与ICC ckit基因水平表达下调引起ICC发育受影响有关。关键词 Cajal间质细胞;ckit;小肠
Exploring the Effect of Blockade of ckit on Interstitial Cells of Cajal and its Mechanisms in the Balb/c Murine Small Intestine

Abstract Objective:To investigate the effect of blockade of ckit on interstitial cells of Cajal in the Balb/c murine small intestine. Methods:Newborn murine were injected intraperitoneally antibody of ckit(ACK2) 100μg every second day, and control groups were injected saline. The distribution of ICC was observed by immunohistochemistry and the expressions of ckit were measured by RTPCR and Western Blot when the murine were 10 days postpartum.Results:The distribution of ICC was lost and the mRNA and protein expression of ckit were significantly lower when ckit were blocked. Conclusion:When the cKit signal is blocked, ICC loss is found and the mechanisms of which may be related with ckit downregulated. ......
